PacBio RS II reagent kits are available in several packaging options emphasizing the granularity and flexibility of our system. You can sequence DNA samples in an individual SMRT Cell or across multiple SMRT Cells according to your experiment design and data needs. This customizable run capability provides for cost efficiencies in both large and small sequencing projects.
The PacBio RS II front-end preparation and on-instrument sequencing workflow is supported by three reagent kits.
Front End Preparation
The DNA template preparation and DNA polymerase binding steps may be performed manually at the benchtop or by an automated liquid-handling system.
1. DNA Template Preparation Reagent Kits
Isolated DNA samples are converted into primer-annealed SMRTbell library format.
2. DNA Polymerase Binding Reagent Kits
Prepared DNA samples, in SMRTbell library format, are bound to the DNA polymerase in preparation for sequencing.
DNA Sequencing
The DNA sequencing step is performed on the PacBio RS II. Sequencing reagent kits supply all materials needed for easy instrument loading.
3. DNA Sequencing Reagent Kit
Supports automated, on-instrument DNA sequencing. Used in conjunction with SMRT Cell 8Pacs.
The DNA Template Prep Kit 2.0 (250 bp - <3 kb) contains reagent supplies to perform DNA template library preparation of primer-annealed SMRTbell libraries for insert sizes less than 3 kb. Reagent quantities support 10 library preparations.
The DNA Template Prep Kit 2.0 (3 kb - < 10 kb) contains reagent supplies to perform DNA template library preparation of primer-annealed SMRTbell libraries for insert sizes greater than 3 kb. Reagent quantities support 10 library preparations.
The DNA/Polymerase Binding Kit 2.0 contains reagent supplies to bind prepared DNA template libraries to the DNA polymerase in preparation for sequencing on the PacBio RS II. Reagent quantities support 24 binding reactions.
The DNA Sequencing Kit 2.0 contains reagent supplies for single molecule real time DNA sequencing on the PacBio RS II instrument to be used in conjunction with SMRTCell 8Pac(s). Reagent quantities support 8 sequencing reactions.
The DNA Control Complex 2.0 (250 bp - <3 kb) contains spike-in template complexes for support of insert sizes from 250 bp to less then 3 kb. Reagent quantities provide spike-in controls for a minimum of 24 samples.
The DNA Control Complex 2.0 (3 kb - 10 kb) contains spike-in template complexes for support of insert sizes equal or greater than 3 kb. Reagent quantities provide spike-in controls for a minimum of 24 samples.
The DNA Sequencing Kit XL 1.0 contains reagent suppplies for single molecule real-time sequencing on the PacBio RS II to be used in conjunction with SMRT Cell 8Pac(s).
The DNA/Polymerase Binding Kit XL 1.0 contains reagent supplies to bind prepared DNA template libraries to the SA-DNA ploymerase in preparation for sequencing on the PacBio RS II. Reagent quantities support 24 binding reactions.
The Plasmidbell Complex XL 1.0 (11 kb) contains a fixed length DNA template of 11 kb bound to SA-DNA ploymerase XL v1.
The MagBeat Kit contains reagent supplies to capture DNA polymerase and template complex for loading onto the PacBio RS II. Kit components include: MagBeads and MagBead Buffer kit.
AMPure PB magnetic beads are specifically formulated for the SMRT sequencing workflow. Beads are used for DNA cleanup and purification during the sample extraction and at various stages in the workflow. Reagent quantities supplied at 60 ml vol.
